Abstract:
An embryo culturing incubator is provided with a shelf configuration that enhances air circulation inside of the incubator, and enables unimpeded access to the individual embryo culture plates that are disposed on the shelves in the incubator. The incubator has an access door which allows one contact with the embryo culture plates on the incubator shelves. The shelves are configured so as to provide a useful free space between the inner surface of the incubator door and the frontal surface of the shelves in the incubator. The size of the free space allows a technician to reach all of the culture plates in the incubator without interfering with other culture plates in the incubator. The free space also allows for exemplary air circulation inside of the incubator.

Abstract:
An apparatus for the culturing of oocytes, embryos, stem cells and cells allows the culturing of the specimens by communally growing or grouping of the specimens and maintains identification of the specimens and allows for the ease in use and location of the specimens in the apparatus. The ability to group the embryos increases cleavage rates, embryos scores and increases the likelihood of better embryos and pregnancy results. This will increase the chances of the survival and future growth or use of the specimens. The apparatus takes the form of a dish which is formed with a plurality of integral wells in which the specimens are placed. The wells or chambers may be subdivided into separate compartments which contain individual embryos or other specimens that are being cultured. The compartments will be interconnected so that the culturing media and culturing byproducts from each specimen can be shared by all of the specimens in any one well, but so that the specimens cannot migrate from one compartment to another.

Abstract:
A biological specimen processing dish assembly includes a dish portion for use in treating specimens such as oocytes, embryos, tissues, and the like sequientially with different reagents or media. The dish includes a plurality of geometrically arranged wells disposed in related sets. The wells are operative to receive reagents or media for the use in treatment of the specimens in question. The specimens can be subjected to a sequence of different treatment media in the wells in a set thereof so as to provide a coherant device for subjecting specimens to a treatment procedure that requires the specimens to be subjected to different media or reagents during sequencial steps in the treatment protocol. The dish can be partially filled with a layer of oil that overlies the reagents in the wells and protects the specimens from exposure to air during the treatment protocol. The dish can be provided with an internal partition that segments the dish so as to provide a plurality of wells that will not be covered by the oil layer. The assembly can also include a cover for overlying the dish so as to provide areas where protocol information can be displayed to enable the technician to properly use the dish assembly in performance of the treatment protocols.

An oocyte and embryo-positioning apparatus enables a plurality of oocytes, embryos or other cell specimens to be held and positioned for injection therein of fluid molecular reagents such as DNA, RNA, protein, cells, spermatozoa and cells; and removal of cells, fragments of cells, cytoplasm, nuclei and cellular organelles. The oocytes and embryos are kept separate from each other in open marked or unmarked well-compartments that are located in the culture container. Each well-compartment comprises one or more finger like structures that can position one oocyte or one embryo. The lanes between the digits of the fingers may be sloped allowing the oocytes or embryo to position at the narrowest and lowest point of the compartment. The method and oocytes and embryo-positioning apparatus of this invention includes a culturing container, such as a Petrie dish, in which the oocytes and embryos are held for micromanipulation. The Petrie dish preferably contains a plurality of the oocyte or embryo-holding finger like-structures which can be positioned in the Petrie dish in a predetermined pattern. For example, the well-compartments can be positioned in the Petrie dish as a strip of well-compartments between the 12 o'clock and 6 o'clock positions, or in any other planned deployment. Marked well-compartments can be identified by letters or numbers or symbols.

A procedure for cryopreservation and efficient post-thaw recovery of a single or few cells, preferably human spermatozoa, their precursor cells or blastomeres, which is achieved by injecting cells into cell-free mammalian zonae pellucidae. The cells may also be of any other type where there are provided a relatively small number of valuable cells which must be retrieved. The zona pellucida may be cryopreserved with the cells contained therein. The method involves a combination of physical micromanipulation procedures and cryoprotectant mediated cryopropreservation. The zonae may be tracked by positioning them into straws between two small air bubbles prior to preservation. There is a high recovery of zonae and sperm within the zonae, and a high fertilization rate of the sperm using ICSI after recovery.
