Abstract:
A catheter for applying ablation energy to biological tissue having biological fluid flowing thereby includes a shaft having a distal-end region defining a tissue-contacting surface and a fluid-contacting surface. A plurality of band electrodes are positioned at the distal-end region of the shaft. A thermally conductive and non-electrically conductive surface covering, covers a portion of each of the band electrodes substantially coincident with the fluid-contacting surface. Each band electrode thereby has at least one masked portion substantially coincident with the fluid-contacting surface and at least one non-masked portion substantially coincident with the tissue-contacting surface. A catheter carries a plurality of electrodes, each with multiple thermal sensors attached and is used to position the electrodes proximal biological tissue. A processor responsive to each of the thermal sensors determines the temperature at the thermal-sensor/electrode junction. A display provides a graphic representation of the temperatures of the thermal-sensor/electrode junctions. An exemplary representation is a bar graph having the temperature of one thermal sensor at one end and the temperature of another thermal sensor at the other end. The length of the bar graph combined with the position of the bar graph relative to a temperature range region provides an indication of the position of the thermal-sensor/electrode junctions relative to the biological tissue. The processor also monitors the spread between the sensor temperatures and compares it to a threshold value. Base on the result of the comparison, the processor controls the power applied to the electrode.

Abstract:
A voltage regulator operable as a voltage follower while a fusible link is closed and in a regulated voltage mode when the fusible link becomes open. The voltage regulator can be formed on monolithic semiconductor chips. Patterned thin films including aluminum and nickel-iron, and aluminum and polycrystalline silicon, comprise the fusible link. With the fusible link closed, the voltage regulator output is an analog of positive polarity variable voltage levels at the regulator input. Systems powered by the voltage regulator are allowed to be programmed until system programming requiring variable voltage levels is complete. Afterwards, a negative polarity voltage is applied to the regulator input causing a large current to pass through the fusible link once the system programming is completed. Current thereby causes the fusible link to become opened and enables the voltage regulator to begin operating at a regulated voltage in response to positive voltage input.

Apparatus and methods are provided for interacting light with particles, including but not limited to biological matter such as cells, in unique and highly useful ways. Optophoresis consists of subjecting particles to various optical forces, especially optical gradient forces, and more particularly moving optical gradient forces, so as to obtain useful results. In biology, this technology represents a practical approach to probing the inner workings of a living cell, preferably without any dyes, labels or other markers. In one aspect, a particle may be characterized by determining its optophoretic constant or signature. For example, a diseased cell has a different optophoretic constant from a healthy cell, thereby providing information, or the basis for sorting. In the event of physical sorting, various forces may be used for separation, including fluidic forces, such as through the use of laminar flow, or optical forces, or mechanical forces, such as through adhesion. Abstract:
A method for detecting the onset of apoptosis in cells using a moving optical gradient includes the steps of exposing at least a portion of the cells to at least one chemical compound, moving the cells and the optical gradient relative to each other so as to cause displacement of at least some of the cells, measuring the displacement of at least a portion of the displaced cells, comparing the measured displacement with the measured displacement of at least one control cell that has not been treated with the at least one chemical compound. The step of comparing the measured displacement of the control and tested cells determines the onset of apoptosis. Methods are also provided for monitoring cells throughout apoptosis.

Abstract:
A method of screening for inhibitors of the Bcr-Abl tyrosine kinase enzyme using a moving optical gradient includes the steps of providing a panel of cell lines having, on average, different copy numbers of the gene that produces the Bcr-Abl tyrosine kinase enzyme, exposing the panel of cell lines with a chemical compound, moving the cells in the panel of cell lines and the optical gradient relative to each other so as to cause displacement of at least some of the cells, measuring the displacement of at least a portion of the displaced cells in each cell line, and comparing the measured displacements with measured displacements from control cells from each cell line that have not been treated with the chemical. The comparison step determines whether the chemical compound is an inhibitor of the Bcr-Abl tyrosine kinase enzyme.

Abstract:
Apparatus and methods are provided for interacting light with particles, including but not limited to biological matter such as cells, in unique and highly useful ways. Optophoresis consists of subjecting particles to various optical forces, especially optical gradient forces, and more particularly moving optical gradient forces, so as to obtain useful results. In biology, this technology represents a practical approach to probing the inner workings of a living cell, preferably without any dyes, labels or other markers. In one aspect, a particle may be characterized by determining its optophoretic constant or signature. For example, a diseased cell has a different optophoretic constant from a healthy cell, thereby providing information, or the basis for sorting. In the event of physical sorting, various forces may be used for separation, including fluidic forces, such as through the use of laminar flow, or optical forces, or mechanical forces, such as through adhesion. Various techniques for measuring the dielectric constant of particles are provided.
