Sequential Solid Phase Immunoassay Including Contact Pad To Limit Conjugate Flow During Manufacture

US Patent:
8501495, Aug 6, 2013
Filed:
Feb 11, 2010
Appl. No.:
12/704147
Inventors:
Kai Ling Yao - Daly City CA, US
Peter Chun - South San Francisco CA, US
Assignee:
Equal Access to Scientific Excellence - South San Francisco CA
International Classification:
G01N 33/558
G01N 33/563
G01N 33/53
C12N 1/34
US Classification:
436514, 436513, 436530, 436810, 435 71, 4352872, 4352877
Abstract:
A sequential solid phase immunoassay and system is disclosed. The immunoassay utilizes the secondary antibody method for the detection of antibodies in a membrane-based test. The system comprises a test strip including a nitrocellulose membrane having an immobilized antigen in a capture zone on the membrane and a stabilized liquid secondary antibody conjugate. The sequential solid phase immunoassay is performed in a sequential manner with the addition of a fluid specimen being followed by the addition of the stabilized liquid secondary antibody conjugate. The sequential procedure using the system includes allowing the fluid specimen containing antibodies specific to the antigen to pass laterally from the test strip first end through the capture zone. The immobilized antigens in the capture zone capture antibodies specific to the antigen. The stabilized liquid secondary antibody conjugate then binds to the captured antibodies and can be detected visually or by a machine or reader.

Neisseria Detection System

US Patent:
5089394, Feb 18, 1992
Filed:
Sep 13, 1989
Appl. No.:
7/406778
Inventors:
Peter K. Chun - South San Francisco CA
Albert E. Chu - Hillsborough CA
Assignee:
E-Y Laboratories, Inc. - San Mateo CA
International Classification:
C12Q 104
C12Q 154
C12Q 137
G01N 33571
US Classification:
435 34
Abstract:
A specimen containing N. lactamica, N. meningitidis, N. gonorrhoeae or B. catarrhalis is incubated simultaneously with a first substrate for beta-galactosidase and a second substrate for gamma-glutamyl aminopeptidase to form reaction products with one or the other Neisseriae which yields a detectible first or second signal distinct from each other depending upon the presence of the enzyme. Signals may comprise distinct colors which emit either in the absence or presence of diazo dye coupler. A third substrate specific for prolyliminopeptidase in N. gonorrhoeae may be added to form a detectible third signal of the same type. The third substrate may be incubated simultaneously with the first two substrates or later. The absence of all of the first three signals is a positive indication that the specimen contains B. catarrhalis.

Directed Flow Diagnostic Device And Method

US Patent:
5006464, Apr 9, 1991
Filed:
Oct 1, 1987
Appl. No.:
7/103845
Inventors:
Albert E. Chu - Hillsborough CA
Peter K. Chun - South San Francisco CA
Siu C. C. Yeung - San Mateo CA
Assignee:
E-Y Laboratories, Inc. - San Mateo CA
International Classification:
G01N 3353
G01N 3122
G01N 33553
US Classification:
435 71
Abstract:
Am improved device and method for analyte assay in liquid samples, wherein a porous membrane with an immobilized receptor which is capable of directly or indirectly binding to the analyte is separated from a body of absorbent material capable of absorbing the liquid sample by a septum capable of substantially separating the porous membrane from the absorbent body while substantially directing the flow of the liquid sample from the porous membrane to the absorbent body.

Hla Typing

US Patent:
5256543, Oct 26, 1993
Filed:
May 10, 1991
Appl. No.:
7/698319
Inventors:
Philippe Pouletty - Atherton CA
Peter Chun - South San Francisco CA
Assignee:
Sangstat Medical Corporation - Menlo Park CA
International Classification:
G01N 33543
G01N 33577
US Classification:
435 794
Abstract:
Physiological samples are typed for HLA-B27 by contacting the sample with an antibody which binds to HLA-B7 and is not cross-reactive between HLA-B7 and B27, separating the sample from any complexes which are formed, and then testing the sample for the presence of HLA-B27 with an antibody which is cross-reactive for HLA-B7 and B27 in a STAT test. Particularly, an enzyme conjugate is used which binds to a membrane allowing for the detection of any HLA-B27 bound to the membrane.

Elongated Membrane Flow-Through Diagnostic Device And Method

US Patent:
5571667, Nov 5, 1996
Filed:
Jul 8, 1993
Appl. No.:
8/089429
Inventors:
Albert E. Chu - Hillsborough CA
Peter K. Chun - South San Francisco CA
Siu Chin C. Yeung - San Mateo CA
International Classification:
C12Q 170
US Classification:
435 5
Abstract:
An apparatus is provided for use in assays for the detection of a bindable target substance in a liquid sample. The apparatus comprises an elongate reaction membrane strip which is porous and liquid-permeable, the upper surface of the membrane strip having receptors thereon capable of binding to the target substance; an absorbent material located adjacent to the lower surface of the reaction membrane strip; and a container means for the reaction membrane strip and absorbent material, including a top wall defining a fluid port in the form of an elongate slot adjacent the membrane strip and including a fluid seal means disposed at the periphery of the fluid port defining a seal between the container top wall and reaction membrane strip. In one embodiment, the receptors on the membrane strip correspond to Western blot proteins. An assay for the detection of bindable target antibody in a liquid sample employing the apparatus is also described.

Hla Typing

US Patent:
5420013, May 30, 1995
Filed:
Aug 2, 1993
Appl. No.:
8/101259
Inventors:
Philippe Pouletty - Atherton CA
Peter Chun - South San Francisco CA
Chin H. Chang - Los Altos CA
Assignee:
Sangstat Medical Corporation - Menlo Park CA
International Classification:
G01N 33543
G01N 33577
US Classification:
435 724
Abstract:
Methods and compositions are provided for detecting HLA polymorphs where distinguishing antibodies are not readily available. Cross-reactive antibodies are employed, where one cross-reacts with the HLA of interest and other alleles, and the other cross-reacts with one of the other alleles, but not with the HLA of interest. By appropriate protocols and controls, the polymorph of interest may be detected.
