Abstract:
A novel function for the p53 gene related to resolution of deep venous thrombosis is disclosed herein. Lack of the p53 gene results in impaired thrombus resolution in a clinically relevant in vivo model of deep venous thrombus resolution. It is further shown that augmentation of p53 activity with quinacrine accelerates thrombus resolution in vivo, and that this beneficial effect is completely dependent on p53. p53-based therapy is therefore provided to accelerate thrombus resolution in patients, and to prevent or ameliorate the debilitating long-tem complications of deep venous thrombosis such as post-thrombotic syndrome.

Abstract:
Disclosed are devices and methods for automated emergency arterial cannulation. The device comprises a hand-held automated emergency arterial sheath placement device having a body with a handle and an actuator, and an arterial sheath placement head at the distal end of the handle body configured for placement against the patient's skin. The arterial sheath placement head includes an arterial sheath insertion device configured to advance an arterial needle, guide wire, and arterial sheath into a patient's artery upon sequential operation of the actuator. The arterial sheath placement head also includes a non-invasive artery locator configured to locate an artery into which the arterial sheath is intended to be placed. The arterial sheath placement head also preferably includes a Doppler gel and antiseptic needle, a local anesthetic needle, and a scalpel, all of which are likewise configured to advance from the upper body portion upon sequential operation of the actuator.

Abstract:
It has now been discovered that p53 is a potent inhibitor of angiogenesis and arteriogensis. Inhibition of p53 stimulates angiogenesis, arteriogenesis and improves perfusion in limbs. Therefore, methods are provided for treating diabetic-induced ischemia in a subject in need thereof comprising locally administering a therapeutically effective amount of one or more p53 molecule inhibitors or analogs and derivatives thereof to a site of ischemic tissue in the subject. Methods are provided for improving limb perfusion in a subject in need thereof comprising locally administering a therapeutically effective amount one or more p53 inhibitors together with a therapeutic agent or separately from the therapeutic agent to cells of a limb of a subject in need thereof. Methods are also provided for improving ischemia-induced angiogenesis and arteriogenesis in method of improving ischemia-induced angiogenesis in tissue of a limb comprising administering a therapeutically effective amount one or more p53 inhibitors individually or in combination with another therapeutic to a subject in need thereof.
