The present invention pertains to compositions and methods for plasmid-mediated supplementation. The compositions and method are useful for retarding the growth of the tumor, and retarding cachexia, wasting, anemia and other effects that are commonly associated in cancer bearing animals. Overall, the embodiments of the invention can be accomplished by delivering an effective amount of a nucleic acid expression construct that encodes a GHRH or functional biological equivalent thereof into a tissue of an animal and allowing expression of the encoded. gene in the animal. For example, when such a nucleic acid sequence is delivered into the specific cells of the tissue specific constitutive expression is achieved.

Electrode Assembly For Constant-Current Electroporation And Use

US Patent:
7245963, Jul 17, 2007
Filed:
Mar 7, 2002
Appl. No.:
10/360768
Inventors:
Ruxandra Draghia-Akli - Houston TX, US
Robert H. Carpenter - Bastrop TX, US
Douglas R. Kern - The Woodlands TX, US
Allan Westersten - Georgetown CA, US
William R. Wilkinson - Scottsdale AZ, US
Assignee:
ADViSYS, Inc. - The Woodlands TX
International Classification:
A61N 1/30
US Classification:
604 20
Abstract:
The present invention relates to a modular electrode system, and its use, for facilitating the introduction of a macromolecule into cells of a selected tissue in a body or plant. The modular electrode system comprises a plurality of needle electrodes; a hypodermic needle; an electrical connector that provides a conductive link from a programmable constant-current pulse controller to the plurality of needle electrodes; and a power source. In a preferred embodiment of the present invention, an operator can grasp the plurality of needle electrodes that are mounted on a support structure and firmly insert the them into the selected tissue in a body or plant. The macromolecules are then delivered via the hypodermic needle into the selected tissue. The programmable constant-current pulse controller is activated and constant-current electrical pulse is applied to the plurality of needle electrodes. The applied constant-current electrical pulse facilitates the introduction of the macromolecule into the cell between the plurality of electrodes.

System And Method For Regulating Pressure Within A Well Annulus

US Patent:
8353351, Jan 15, 2013
Filed:
May 20, 2010
Appl. No.:
12/784367
Inventors:
Robert B. Carpenter - Tomball TX, US
Omid Oujani - Houston TX, US
Krystian K. Maskos - Houston TX, US
John Lofton - Woodlands TX, US
Assignee:

International Classification:
E21B 21/00
US Classification:
166338, 166344, 166347, 166351, 166311
Abstract:
Pressure within a subsea well is managed as temperature within the well fluctuate. The management of the pressure mitigates stress to the structure of the well caused by the pressure. To manage the pressure, fluid is received from and/or provided to the well to reduce and/or increase pressure within the well.

US Classification:
604 21, 604 20
Abstract:
An electroporation device which may be used to effectively facilitate the introduction of a macromolecule into cells of a selected tissue in a body or plant. The electroporation device comprises an electro-kinetic device (“EKD”) whose operation is specified by software or firmware. The EKD produces a series of programmable constant-current pulse patterns between electrodes in an array based on user control and input of the pulse parameters and allows the storage and acquisition of current waveform data. The electroporation device also comprises a replaceable electrode disk having an array of needle electrodes, a central injection channel for an injection needle, and a removable guide disk.

Administration Of Nucleic Acid Sequence To Female Animal To Enhance Growth In Offspring

US Patent:
2003007, Apr 17, 2003
Filed:
Dec 12, 2001
Appl. No.:
10/021403
Inventors:
Robert Schwartz - Houston TX, US
Ruxandra Draghia-Akli - Houston TX, US
Roy Smith - Houston TX, US
Douglas Kern - The Woodlands TX, US
Robert Carpenter - Bastrop TX, US
International Classification:
A01K067/027
C12N015/88
US Classification:

Abstract:
Growth is improved by utilizing growth enhancement potential methodology to administer a nucleic acid sequence, such as GHRH or an analog, to a female animal, preferably through a parenteral route of administration. Piglets born from sows injected with DNA encoding GHRH are larger, and effects are demonstrated in subsequent pregnancies without additional administration(s) of the vector.

Increased Delivery Of A Nucleic Acid Construct In Vivo By The Poly-L-Glutamate ( Plg ) System

US Patent:
2004001, Jan 22, 2004
Filed:
Mar 24, 2003
Appl. No.:
10/395709
Inventors:
Ruxandra Draghia-Akli - Houston TX, US
Robert Carpenter - Bastrop TX, US
Douglas Kern - The Woodlands TX, US
Leigh Hill - Houston TX, US
Heather Attra - Houston TX, US
Henry Hebel - The Woodlands TX, US
Assignee:
ADVISYS, Inc. - The Woodlands TX
International Classification:
A61K048/00
C12N015/85
US Classification:
514/008000, 514/044000, 435/455000, 435/320100
Abstract:
Plasmid DNA delivered by injection/electroporation to the skeletal muscle can be expressed, and physiologic levels of transgene could be achieved into the circulation. Nevertheless, stabilization of naked DNA may be required and necessary in some cases, as prolonged storage at different temperatures before usage, injection into a large number of animals, etc. It is imperative that the associated compound should not be toxic to the cells (e.g. muscle cells) or cause breakage of plasmid DNA. It would be preferable for the coated DNA to have a similar or increased uptake into the target cells. Low molecular weight poly-L-glutamate compounds have all the desired properties. It was determined that mole/mole ratio DNA/PLG is the optimum concentration for gene therapeutic applications to the skeletal muscle, resulting in increased expression of the transgene, with no damage to the target tissue. Furthermore, stabilization of plasmid DNA by PLG has never been observed or described in the literature.
