Ceramide Anionic Liposome Compositions

US Patent:
2012028, Nov 15, 2012
Filed:
May 10, 2012
Appl. No.:
13/468578
Inventors:
Mark Kester - Harrisburg PA, US
Todd Fox - Hershey PA, US
Assignee:
The Penn State Research Foundation - University Park PA
International Classification:
A61K 31/475
B01J 13/04
A61K 31/7076
A61P 35/00
A61K 9/127
A61K 31/44
US Classification:
424450, 514283, 514350, 514 46, 264 41
Abstract:
Described herein are pharmaceutical compositions according to aspects of the present invention which include one or more hydrophilic antineoplastic chemotherapeutics, such as vinca alkyloid antineoplastic chemotherapeutics, encapsulated in ceramide anionic liposomes. Methods of treatment of a subject having cancer using the pharmaceutical compositions are described, along with methods of making ceramide anionic liposomes which encapsulate one or more hydrophilic antineoplastic chemotherapeutics in the aqueous interior of the ceramide anionic liposomes.

Method And System For Altering Dysfunctional Lipid Metabolism In Diabetic Complications

US Patent:
2008013, Jun 5, 2008
Filed:
Jul 20, 2007
Appl. No.:
11/880446
Inventors:
Mark Kester - Harrisburg PA, US
Todd E. Fox - Hershey PA, US
International Classification:
A61K 31/5375
A61P 27/02
US Classification:
5142378
Abstract:
Diabetic retinopathy is a debilitating complication of diabetes and a leading cause of vision loss, however the fundamental mechanisms contributing to vision loss remain undefined. Several novel observations are described: 1) diabetic retinas demonstrate decreased total ceramide levels; 2) with a concomitant increase in glucosylceramides; 3) which mediates decreased insulin receptor signaling and; 4) cell death in vitro and in vivo. Inhibition of this dysfunctional glycosphingolipid metabolism restores insulin sensitivity. Moreover, elevation in diglycerides and reduction in concentrations of phosphatidic acid or ceramide-1-phosphate also contribute to diabetic complications and insulin resistance. The mechanism responsible for dysfunctional lipid metabolism in diabetic tissues involves reduced caveloin--expression within structured membrane microdomains and suggest another target for molecular and pharmacological intervention. Preferred embodiments describe pharmacological and molecular systems and methods to therapeutically alter dysfunctional lipid metabolism and restore selective insulin-dependent kinase cascades as well as membrane integrity.

Dhs1P And Use Of Same As An Anticancer Therapeutic And Immunomodulator

US Patent:
2016020, Jul 21, 2016
Filed:
Jan 15, 2016
Appl. No.:
14/997242
Inventors:
- University Park PA, US
Mark Kester - Harrisburg PA, US
James H. Adair - State College PA, US
Todd E. Fox - Hershey PA, US
International Classification:
A61K 31/661
A61K 45/06
A61K 41/00
A61K 9/48
A61K 35/13
Abstract:
Use of dhS1P and/or PhotoImmunoNanoTherapy as a therapeutic agent is described. Administration of therapeutically effective amounts of dhS1P decrease the number of Myeloid Derived Suppressor Cells and immune suppression in cancer patients. Administration of therapeutically effective amounts of dhS1P can be used as an adjuvant to conventional cancer therapies including immunotherapies. Therapeutic results can be achieved by directly administering dhS1P and/or by indirectly increasing the amount of dhS1P at the tumor site. The therapy permits the patient's immune system to recognize and eliminate cancer cells reducing tumor size and extending patient survival.
